- - ใช้รักษาโรคเริมที่เกิดจาก herpes simplex virus ที่ผิวหนังและเยื่อบุเมือกรวมทั้งเริมที่อวัยวะเพศที่เป็นครั้งแรกและที่กลับมาเป็นซ้ำอีก (initial and recurrent genital herpes) - ใช้ป้องกันโรคเริมที่กลับมาเป็นซ้ำอีกในผู้ป่วยภูมิคุ้มกันปกติและป้องกันการเกิดโรคเริมในผู้ป่วยภูมิคุ้มกันบกพร่อง - ใช้รักษาโรคอีสุกอีใส (varicella infections, Chickenpox) - ใช้รักษาโรคงูสวัด (herpes zoster infections, Shingles) - ป้องกันการติดเชื้อ CMV ในผู้ป่วยที่ได้รับการปลูกถ่ายไขกระดูก พบว่าการให้ Acyclovir ชนิดหยดเข้าหลอดเลือดดำเป็นเวลา 1 เดือน ตามด้วย Acyclovir ชนิดรับประทานเป็นเวลา 6 เดือน ช่วยลดอัตราการเสียชีวิตในผู้ป่วยที่ได้รับการปลูกถ่ายไขกระดูกได้
